چکیده

Abstract Background As malaria incidence and transmission in a region decreases, it becomes increasingly difficult to identify areas of active transmission. Improved methods for identifying monitoring foci are needed low parasite prevalence order achieve elimination. Serological assays can provide population-level infection history inform elimination campaigns. Methods A bead-based multiplex antibody detection assay was used evaluate chimeric Plasmodium vivax MSP1 protein (PvRMC-MSP1), designed be broadly immunogenic use vaccine studies, act as pan-malaria serological tool based on its ability capture IgG plasma samples obtained from naturally exposed individuals. Samples 236 US travellers with PCR confirmed status all four major species infecting humans, falciparum (n = 181) , 38), malariae 4), ovale 13) were tested using PvRMC-MSP1 well the recombinant MSP1-19 kD isoforms representative these species. Results Regardless species, large proportion infected provided high signal protein, 115 responders out assessed (48.7%). When grouped by infection, 38.7% P. falciparum- 92.1% vivax- 75.0% malariae- 53.4% ovale- individuals displayed signals response PvRMC-MSP1. It also determined that -infected produced increased chimera compared PvMSP1 89.5% (34 38) showed improved antibodies when PvMSP1, observed just 1.1% who protein. Conclusions These results support further study antigens an approach increasing sensitivity or broadening binding capacity improve existing tools determining exposure Including both broad-reacting species-specific antigen-coated beads panel could nuanced view histories, extensive profile, detailed seroestimates. more sensitive would aid campaigns co-endemic regions where is dominant parasite.

Antibodies to Plasmodium falciparum and Plasmodium vivax merozoite surface protein 5 in Indonesia: species-specific and cross-reactive responses.

BACKGROUND Merozoite surface protein (MSP) 5 is a candidate antigen for a malaria vaccine. In cross-sectional and longitudinal studies, we measured MSP5 antibody responses in Papuans with acute Plasmodium falciparum malaria, Plasmodium vivax malaria, and mixed P. falciparum and P. vivax malaria and in those with past exposure. METHODS Enzyme-linked immunosorbant assay (ELISA) was used to quan...

Induction of Multifunctional Broadly Reactive T Cell Responses by a Plasmodium vivax Circumsporozoite Protein Recombinant Chimera.

Plasmodium vivax is the most widespread species of Plasmodium, causing up to 50% of the malaria cases occurring outside sub-Saharan Africa. An effective vaccine is essential for successful control and potential eradication. A well-characterized vaccine candidate is the circumsporozoite protein (CSP). Preclinical and clinical trials have shown that both antibodies and cellular immune responses h...

Gametocytemia in Plasmodium vivax and Plasmodium falciparum infections.

Two expert research microscopists, each blinded to the other's reports, diagnosed single-species malaria infections in 2,141 adults presenting at outpatient malaria clinics in Tak Province, Thailand, and Iquitos, Peru, in May-August 1998, May-July 1999, and May-June 2001. Plasmodium vivax patients with gametocytemia had higher fever and higher parasitemia than those without gametocytemia; tempe...

Stage-specific and species-specific antigens of Plasmodium vivax and Plasmodium ovale defined by monoclonal antibodies.

Monoclonal antibodies (MAbs) were produced against the asexual blood stages of Plasmodium vivax and Plasmodium ovale and used to define antigens of plasmodial parasites in an indirect fluorescent antibody assay. The anti-P. vivax MAbs produced two distinct patterns in the indirect fluorescent antibody assay. Four patterns were found with the anti-P. ovale MAbs. Species-specific epitopes were de...

Identification of Protein Markers in Patients Infected with Plasmodium knowlesi, Plasmodium falciparum and Plasmodium vivax

Malaria is caused by parasitic protozoans of the genus Plasmodium and is one of the most prevalent infectious diseases in tropical and subtropical regions. For this reason, effective and practical diagnostic methods are urgently needed to control the spread of malaria. The aim of the current study was to identify a panel of new malarial markers, which could be used to diagnose patients infected...
